研究概述
倪其焜博士為質譜技術專家,其研究團隊認知到構成生命的巨分子之一 : 碳水化合物,至近期仍沒有一個快速有效的初級結構的鑑定方式,因此從2017 年起全力投入這個重要課題的研究。構成生命的三類巨分子 : 核糖核酸,蛋白質,及碳水化合物,至今只有碳水化合物依然沒有可靠快速的方法可以決定其初級結構。碳水化合物初級結構的鑑 定,傳統上是使用各種化學反應法和酵素,或是使用核磁共振光譜,但這兩種方法都耗時、耗樣品。 質譜儀有很高的靈敏度,所需的樣品非常少,但是傳統質譜法只能決定碳水化合物醣酐鍵的位置,無法決定變旋異構物、單醣的立體結構等。 倪博士實驗室研究團隊,結合實驗,量子化學計算,和化學動力學理論,研究碳水化合物在氣相的分解機制,並進一步利用這些分解機制,提出一個全新的質譜法,名為「邏輯演繹序列串聯質譜法」,可以鑑定醣酐鍵的位置、變旋異構物、單醣的立體結構等。 倪博士實驗室開發的方法,是目前全世界所有質譜法中,唯一可以鑑定寡糖完全初級結構的質譜方法。最近倪博士團隊將這個方法應用在各種生物體中,發現了前所未見的新寡糖。
